Overview
Gonadorelin is the synthetic form of gonadotropin-releasing hormone (GnRH), the ten-residue peptide that hypothalamic neurons release into the pituitary portal circulation to control reproductive hormone output. Its structure, pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, was worked out in the early 1970s by the laboratories of Andrew Schally and Roger Guillemin, work recognised with a share of the 1977 Nobel Prize in Physiology or Medicine. Because it is identical to the native hormone, gonadorelin has long served as a reference ligand in reproductive endocrinology.
Gonadorelin is an approved prescription product in several jurisdictions for specific diagnostic indications, notably assessment of pituitary gonadotroph reserve. In the laboratory it is central to one of endocrinology's best-known observations: the pituitary responds very differently to pulsatile versus continuous GnRH exposure. That finding shaped the development of long-acting GnRH agonists and antagonists and continues to inform research on puberty, hypothalamic amenorrhoea and the neural pulse generator. Gonadorelin is supplied here strictly as a research reference material.
Mechanism studied
- Gonadorelin
- Pituitary GnRH receptor
- LH & FSH
- Gonads
Gonadorelin binds the GnRH receptor (GnRHR), a class A G protein-coupled receptor on pituitary gonadotrophs that is unusual in lacking a cytoplasmic C-terminal tail. Receptor activation couples primarily to Gq/11, stimulating phospholipase C, inositol trisphosphate generation, intracellular calcium mobilisation and protein kinase C activity. These signals drive synthesis and release of luteinising hormone (LH) and follicle-stimulating hormone (FSH), which in turn act on the gonads. Pulse frequency matters: faster pulses favour LH beta-subunit transcription, slower pulses favour FSH beta. Sustained, non-pulsatile exposure produces receptor desensitisation and uncoupling, suppressing gonadotropin output, a phenomenon studied extensively in pituitary cell and primate models.
Research highlights
- 01Isolated and sequenced in 1971 from porcine and ovine hypothalami; Schally and Guillemin shared part of the 1977 Nobel Prize for hypothalamic hormone research.
- 02Ernst Knobil's rhesus monkey experiments in the late 1970s showed that intermittent GnRH sustains gonadotropin secretion while continuous exposure suppresses it.
- 03Pituitary desensitisation research with gonadorelin underpinned the design of later GnRH superagonists and antagonists.
- 04Used as a stimulation reference in studies of delayed puberty, central precocious puberty and hypogonadotropic hypogonadism.
- 05Open questions include how pulse frequency is decoded at the transcriptional level and how upstream kisspeptin neurons set the pulse generator.

FAQ
Yes. Gonadorelin has the same ten-amino-acid sequence as mammalian GnRH-I, including the N-terminal pyroglutamate and C-terminal glycinamide. This identity is why it is used as a reference ligand, whereas analogues such as leuprorelin or triptorelin carry substitutions that alter stability and receptor kinetics.
Pituitary gonadotrophs are tuned to intermittent signals. Pulsatile GnRH maintains LH and FSH secretion, while continuous exposure leads to receptor desensitisation and suppression. This dual behaviour is a foundational concept in reproductive endocrinology and explains why research designs carefully control the timing of exposure.
The World Anti-Doping Agency lists GnRH and its analogues under section S2 (peptide hormones, growth factors and related substances), prohibited in males because they can raise testosterone. Researchers in sports science should consult the current WADA Prohibited List.
