Vasopressin (ADH) analogue with antidiuretic and haemostatic activity

Reference compound — not available from Purity Peptides. Shown for research context.

Overview

Desmopressin, chemically 1-deamino-8-D-arginine vasopressin, is a synthetic analogue of the posterior pituitary hormone vasopressin. It was first described in the late 1960s by Czech chemists, who removed the N-terminal amino group and replaced L-arginine with D-arginine at position 8. These two changes extend its duration of action and shift its selectivity strongly toward the V2 receptor, largely separating antidiuretic activity from the pressor effects of the parent hormone.

Desmopressin is approved as a prescription medicine for indications including central diabetes insipidus, nocturia and certain bleeding disorders such as mild haemophilia A and von Willebrand disease. In research, it is the standard selective V2 agonist for studying renal water handling, aquaporin-2 trafficking and the regulation of plasma sodium. Its ability to release von Willebrand factor and factor VIII from endothelial stores has made it an important probe in haemostasis research. Hyponatraemia, arising from excessive water retention, is the most studied safety concern.

Mechanism studied

  1. Desmopressin
  2. Renal V2 receptor
  3. Aquaporin-2 insertion
  4. Water reabsorption

Desmopressin is a selective agonist of the vasopressin V2 receptor, a Gs-coupled GPCR on the basolateral membrane of renal collecting duct principal cells. Receptor activation raises cyclic AMP, activates protein kinase A and drives phosphorylation and insertion of aquaporin-2 water channels into the apical membrane. Water then moves from the tubular fluid back into the circulation, concentrating the urine. V2 receptors on vascular endothelium mediate a separate effect: release of von Willebrand factor and factor VIII from Weibel-Palade bodies. Desmopressin's minimal V1a activity explains its weak vasoconstrictor action compared with native vasopressin.

Research highlights

  • 01First synthesised in the late 1960s through targeted modification of vasopressin's N-terminus and position 8.
  • 02Became the archetypal selective V2 agonist for dissecting renal water reabsorption and aquaporin-2 biology.
  • 03Discovery of its effect on von Willebrand factor release opened a distinct line of haemostasis research.
  • 04Studied in nocturia and enuresis research, where age-related sensitivity to hyponatraemia is a key variable.
  • 05Open questions include individual variation in response and better predictors of water-retention risk.

FAQ

  • Desmopressin lacks the N-terminal amino group and contains D-arginine at position 8. These changes slow its breakdown and make it highly selective for V2 receptors, so it retains antidiuretic activity while having little pressor effect.

  • Activation of endothelial V2 receptors releases von Willebrand factor and factor VIII from storage granules. Researchers use desmopressin to study this release pathway and its role in mild inherited bleeding disorders.

  • Desmopressin is an approved prescription medicine in the US and EU for specific indications. WADA lists it as a prohibited masking agent because it can alter urine concentration. Research material is for laboratory use only.
