Dual GLP-1 and glucagon receptor agonist

Reference compound — not available from Purity Peptides. Shown for research context.

Overview

Survodutide is an investigational dual agonist of the glucagon and GLP-1 receptors developed by Boehringer Ingelheim in collaboration with Zealand Pharma. It is an acylated peptide with a long half-life suitable for once-weekly regimens in clinical trials, and its design reflects the hypothesis that adding glucagon-receptor activity to GLP-1 agonism can increase energy expenditure and directly reduce liver fat.

A phase 2 trial in people with metabolic dysfunction-associated steatohepatitis (MASH) and fibrosis, published in 2024, reported improvement in MASH without worsening fibrosis in a large majority of participants receiving the compound. Phase 2 obesity studies also reported meaningful weight reductions. Phase 3 programmes in obesity (SYNCHRONIZE) and MASH are ongoing. Survodutide is not approved anywhere. Our product is a research-grade reference compound for studies of hepatic metabolism and dual-receptor pharmacology.

Mechanism studied

  1. Survodutide
  2. GLP-1R + GCGR
  3. Hepatic lipid oxidation
  4. Liver & weight endpoints

Survodutide activates the glucagon receptor, abundant in the liver, and the GLP-1 receptor, found in the pancreas, gut and brain. Hepatic glucagon-receptor signalling increases fatty-acid oxidation and reduces lipogenesis, lowering liver fat, and may raise energy expenditure. GLP-1 receptor signalling reduces appetite, slows gastric emptying and enhances glucose-dependent insulin release, offsetting the glucose-raising potential of glucagon agonism. The relative balance between the two activities is a key design parameter studied across this drug class.

FAQ

  • Glucagon receptors are highly expressed in the liver, and activating them increases fat oxidation in hepatocytes. Combining this with GLP-1 agonism has made survodutide a candidate in MASH research, where reductions in liver fat and inflammation are key endpoints in clinical and preclinical studies.

  • Lyophilized Survodutide is normally kept sealed at −20 °C, protected from light and moisture. Once reconstituted for laboratory work, aliquots are held at 2–8 °C for short-term use or frozen, avoiding repeated freeze–thaw. Follow the lot-specific guidance on the certificate of analysis.

  • Every lot of Survodutide ships with a certificate of analysis reporting HPLC purity, mass-spectrometry identity against the expected molecular weight, appearance and lot number. Match the COA lot to the vial label and keep it with your experimental records.
