Potent long-acting GnRH agonist delivered by implant

Reference compound — not available from Purity Peptides. Shown for research context.

Overview

Histrelin is a synthetic nonapeptide analogue of gonadotropin-releasing hormone and one of the most potent GnRH agonists described. It carries a benzyl-protected D-histidine at position 6 and a C-terminal proline ethylamide, modifications that markedly increase receptor affinity and resistance to degradation. In the United States it is marketed in a long-duration hydrogel reservoir formulation as Supprelin LA for central precocious puberty, and previously as Vantas for advanced prostate cancer.

Histrelin is of interest to researchers studying continuous, near-constant GnRH receptor exposure, because its reservoir design releases peptide steadily over many months. This allows investigation of long-term pituitary suppression without the peaks and troughs associated with shorter formulations. Studies of central precocious puberty have used it to examine how suppression of the reproductive axis affects growth velocity, skeletal maturation and later recovery of pubertal development. It also contributes to the broader comparative literature on how GnRH agonist potency and release kinetics relate to the depth of sex-steroid suppression.

Mechanism studied

  2. GnRH receptor
  3. Continuous suppression
  4. Sex steroids ↓

Histrelin binds GnRH receptors on anterior pituitary gonadotrophs with very high affinity. As with other superagonists, early exposure stimulates luteinising hormone and follicle-stimulating hormone release. Continuous release from its reservoir formulation then maintains uninterrupted receptor occupancy, producing desensitisation, receptor internalisation and suppressed gonadotropin synthesis. The outcome is sustained reduction of gonadal sex-steroid production for the life of the formulation. Because suppression depends on ongoing receptor exposure, it is reversible: reproductive axis activity resumes after the formulation is removed, a feature examined in recovery studies in central precocious puberty research.

Research highlights

  • 01Developed through position-6 substitution work in the 1980s, yielding one of the highest-affinity GnRH agonists known.
  • 02Its hydrogel reservoir formulation is a notable example of long-duration peptide release engineering.
  • 03Studied extensively in central precocious puberty, including research on post-suppression recovery of the axis.
  • 04Previously studied and marketed for advanced prostate cancer under a separate brand name.
  • 05Open questions concern long-term growth, bone and metabolic outcomes after prolonged pubertal suppression.

FAQ

  • The benzyl-protected D-histidine at position 6 and the C-terminal ethylamide substantially raise histrelin's affinity for the GnRH receptor and slow its breakdown, placing it among the most potent GnRH agonists characterised in receptor studies.

  • Histrelin is supplied clinically in a hydrogel reservoir that releases the peptide steadily over many months. This provides near-constant receptor exposure and has been studied as a model of long-duration peptide release.

  • Histrelin is an approved prescription medicine in the United States for central precocious puberty, and a prostate cancer product was previously marketed. Registration in other regions varies. Research material is for laboratory use only.
